Background:IL33 is a cytokine which belongs to the IL-1 superfamily, and it induces helper T cells to produce type 2 cytokines. This cytokine was previously named NF-HEV 'nuclear factor (NF) in high endothelial venules' (HEVs), as it was originally identified in these specialized cells.
IL33 mediates its biological effects by interacting with the receptors ST2 and IL-1 Receptor Accessory Protein, activating intracellular molecules in the NF-kappaB and MAP kinase signaling pathways that drive production of type 2 cytokines (e.g. IL-4, IL-5 and IL-13) from polarized Th2 cells. The induction of type 2 cytokines by IL-33 in vivo is believed to induce the the severe pathological changes observed in mucosal organs following administration of IL33.

VALIDATION IMAGES

Tissue/cell: rat stomach tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer ( 0.01M, pH 6.0 ), Boiling bathing for 15min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30min; Blocking buffer (normal goat serum,C-0005) at 37℃ for 20 min;
Incubation: Anti-IL33 Polyclonal Antibody, Unconjugated(bs-2633R) 1:200, overnight at 4°C, followed by conjugation to the secondary antibody(SP-0023) and DAB(C-0010) staining

Paraformaldehyde-fixed, paraffin embedded (Rat colon); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (IL33) Polyclonal Antibody, Unconjugated (bs-2633R) at 1:400 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.
