Background:DEPTOR (DEP domain containing MTOR-interacting protein), also known as DEP.6 or DEPDC6 (DEP domain-containing protein 6), is a 409 amino acid protein that negatively regulates mTORC1 and mTORC2 pathways. DEPTOR interacts with FRAP via its PDZ domain, and undergoes post-translational phosphorylation. Containing two DEP domains and one PDZ (DHR) domain, DEPTOR is encoded by a gene that maps to human chromosome 8q24.12. Chromosome 8 consists of nearly 146 million base pairs, encodes over 800 genes and is associated with a variety of diseases and malignancies. Schizophrenia, bipolar disorder, Trisomy 8, Pfeiffer syndrome, congenital hypothyroidism, Waardenburg syndrome and some leukemias and lymphomas are thought to occur as a result of defects in specific genes that map to chromosome 8.

VALIDATION IMAGES

Paraformaldehyde-fixed, paraffin embedded (mouse pancreas); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (DEPTOR) Polyclonal Antibody, Unconjugated (bs-8255R) at 1:200 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.

Tissue/cell: rat brain tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer ( 0.01M, pH 6.0 ), Boiling bathing for 15min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30min; Blocking buffer (normal goat serum,C-0005) at 37℃ for 20 min;
Incubation: Anti-DEPTOR Polyclonal Antibody, Unconjugated(bs-8255R) 1:500, overnight at 4°C, followed by conjugation to the secondary antibody(SP-0023) and DAB(C-0010) staining
