Background:Ago1 belongs to the argonaute family and contains 1 PAZ and 1 Piwi domain. It is required for G1 arrest and mating in response to nitrogen starvation. It is also known as cell cycle control protein through it's regulation of cytokinesis and cell cycle checkpoints occurs downstream of dcr1. It is required, indirectly, for regulated hyperphosphorylation of cdc2. Ago 1 also has a role in the RNA interference (RNAi) pathway which is important for heterochromatin formation and accurate chromosome segregation. It has been shown to be non-essential for siRNA-directed target RNA cleavage but is needed for the production of mature miRNA that affects miRNA-directed RNA cleavage.

VALIDATION IMAGES

Tissue/cell: mouse brain tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer ( 0.01M, pH 6.0 ), Boiling bathing for 15min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30min; Blocking buffer (normal goat serum,C-0005) at 37℃ for 20 min;
Incubation: Anti-EIF2C Polyclonal Antibody, Unconjugated(bs-7737R) 1:200, overnight at 4°C, followed by conjugation to the secondary antibody(SP-0023) and DAB(C-0010) staining
