Background:Apoptosis inhibitor 5 (API5) is an antiapoptotic factor which may have a role in protein assembly. API5 expression is often upregulated in tumor cells, particularly in metastatic cells. It is a critical determinant of E2F1-induced apoptosis, acting downstream of E2F to suppress E2F-dependent apoptosis without generally blocking E2F-dependent transcription. Function : Antiapoptotic factor that may have a role in protein assembly. Negatively regulates ACIN1. By binding to ACIN1, it suppresses ACIN1 cleavage from CASP3 and ACIN1-mediated DNA fragmentation. Also known to efficiently suppress E2F1-induced apoptosis. Its depletion enhances the cytotoxic action of the chemotherapeutic drugs.

VALIDATION IMAGES

Paraformaldehyde-fixed, paraffin embedded (rat thymus); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (API5) Polyclonal Antibody, Unconjugated (bs-22073R) at 1:400 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.
