Background:The primary purpose of all hot start PCR methods is to prevent Taq DNA polymerase activity prior to thermal cycling. Even if set-up is conducted on ice, the Taq DNA polymerase remains active and may elongate unwanted products such as misprimed or other nonspecific events. Primer-dimer interactions may also be amplified, which will reduce overall yield and efficiency.
